CellGlow Viability Assay Protocol

The CellGlow AO/PI Viability Assay streamlines cell counting by replacing liquid reagents with precision-coated, surface-stabilized fluorophores in a strip-tube format.

CellGlow is compatible with CellDrop FLi, CellDrop FLxi and slide-based fluorescence slide counters equipped with excitation and emission hardware optimized for Acridine Orange and Propidium Iodide. 

  1. Ensure that the cell suspension is well-mixed and aspirate 20 µL.
  2. Pierce the seal of CellGlow well using the pipette tip and dispense the sample to the bottom of the tube. 
    • Note: Ensure that the sample is fully collected at the bottom of the well.
  3. Mix the solution 2 – 3 times. No incubation time is required.

  1. Immediately after mixing, aspirate 10 µL of cell suspension from the well and load into the CellDrop Automated Cell Counter chamber or slide.
    • Note: A dilution factor of 1 should be used for un-diluted samples when using CellGlow.
    • When using liquid-based Viability Assays the dilution factor default is 2 as assay preparation uses a 1:1 volume of dye and cell suspension. There is no dilution effect with CellGlow.

  1. Each tube strip can be torn away for disposal after use. 
  2. Storage and Shelf-Life:
    • Keep at room temperature
    • Replace after 12 months
