CellGlow Viability Assay Protocol

The CellGlow AO/PI Viability Assay streamlines cell counting by replacing liquid reagents with precision-coated, surface-stabilized fluorophores in a 96 well format.

Review this protocol in full before using CellGlow with the CellDrop Automated Cell Counter or any slide-based cell counter.

  1. Ensure that cells are well-mixed, then aspirate 20 µL of cell suspension.
  2. In a single motion, pierce one of CellGlow’s individually sealed wells using the pipette tip, then dispense the sample to the bottom of the assay well. 
    • Note: Ensure that the sample is fully collected at the bottom of the well.
  3. Mix the solution 2 – 3 times. No incubation time is necessary.

  1. Immediately after mixing, aspirate 10 µL of cell suspension from the well and load onto the CellDrop Automated Cell Counter or any slide-based automated cell counter.
    • Note: For the CellGlow Viability Assay, a dilution factor of 1 should be used for un-diluted samples. When using liquid-based DeNovix CellDrop Viability Assays on the CellDrop Automated Cell Counter, the dilution factor default is 2 as assay preparation uses a 1:1 volume of dye and cell suspension.

  1. When all wells have been used within a column, each tube strip can be torn away for disposal. Remove CellGlow from the tray to easily remove a strip.
